My brother-in-law lives in the US and owns land in Thondamuthur. He wants to gift this property to his brother in India. Can he do this without being present in the Registrar’s office? Can he use a Power of Attorney instead? If so, will he need to provide a “live certificate” if the Power of Attorney is more than a month old?

Yes, your brother-in-law can gift the property to his brother in India without being physically present in the Registrar’s office by using a Power of Attorney.
